My practice/back-up amp is a Peavey Classic 30 112 (w/Eminence Private Jack). I use an overdrive in front (MXR Classic OD) to effectively give me 4 channels. It sounds great when engaged over the drive channel but when engaged over the clean channel there is a notable volume drop. What's going on here?

I could be wrong but I think you could have a faulty preamp tube.

Does the clean channel volume drop without the boost?

The boost on the peavey is off. If I change channels without my OD on it's normal. The volume drop occurs while on the clean channel and I engage the OD while engaging the OD while on the drive channel is fine.

Sounds like you need to tweak your volume level on your pedal then. It has more impact on the clean channel than it will on the OD channel.

Even if I crank the vol on the OD I still get a volume drop (somewhat lessoned) but then if I have the gain on the OD up where I want it to effect the clean channel I get WAY too much drive on the drive channel setting.

I know this amp very well as I've owned and modded a few in my life.

Question - the amp runs perfectly fine on its OWN clean and on its OWN dirty (overdrive) correct?

So you've added a stomp in front which effectively offers its 2 types of ON-OFF, therefore allowing you to the following:
OD-off / Peavey Clean
OD-on / Peavey Clean
OD-off / Peavey Dirty
OD-on / Peavey Dirty

Quote this message and tell be on the above combos, where this volume occurs.

I also find that the clean (OD-off) is VERY punchy. Comparatively, I run the clean volume on 1 1/2 and the drive channel gain-6, volume-6.

Alright, the clean on the C30 is always more punchy than the push-button overdrive section. It's actually one of the sweetest cleans to gritty channels out there for the price. Gorgeous tone (try installing a Celestion Alnico Gold in that amp for an AMAZING sound!!).

So you run the overdrive into the clean. With the overdrive off, it's punchy, loud, ballsy, and has that OOOMPH, ya? Then you step ON your overdrive punch WHILE on the Clean Channel and it goes quiet?? What's the stomp you're using? And the stomp is just infront of the amp, yes? Not in that gimmicky loop, correct?

Lemme know.

Using a MXR classic OD in front. Just fyi, I've got an Eminence RedCoat in there that is a HUGE improvement over the (ack)Blue Marvel.
