Is a speaker swap going to change the amp tone?

LOL at the tongue in cheek comments from people insuating the OP is wrong based on his word choice.

The speaker DOES change the AMPLIFIERS' tone as the resulting (changed) impedence load on the ouput section changes the response of said output section amongst other factors.

I'll let you know how much speakers change amp tone when my EVH 4x12 arrives tomorrow :D

I feel like cabs change tone just as much if not more. The reason I say this is different internal reflections/space/speaker codes. I had a VHT fatbottom 2x12 and put webers in it. Not much different tone than the stock eminences. Same with my Splawn 4x12. Swapped some speakers, didn't change that much.

But plugging in to a splawn 2x12 sounds entirely different that a VHT or Mesa 2x12. I had all three at once.

The EVH 4x12 sounded totally different than my splawn 4x12.

Every individual speaker and every individual cab sounds different. The cab has as much effect on the sound as the speaker.
