Anyone ever notice a difference in tone when wiring a 4x12 cabinet parallel/series vs series/parallel?

Yes. However, there is a quite a bit of confusion on the web on what is series/parallel and what is parallel/series. For example.....google image "series/parallel 4x12" and you will see both variations.

Here is what I found with multiple heads using the same cab and speakers. I'll use different terms for clarification sake:

Speaker pairs wired in series/then paralleled at jack:
More lower mids, a touch more sag, slightly quieter

Speaker pairs wired parallel/then serial at jack (uses one more wire):
Brighter, tighter, edgier, and slightly louder

I prefer the former.......especially with Marshally circuits.

I've experimented many times and the difference is not subtle. It's pretty obvious to my ears.

BTW......I also prefer 8 ohm speakers to 16 ohm. Therefore, I wire my 4x12's for 8 ohm mono. I find 8 ohm speakers less fizzy and more focused than their 16 ohm counterparts. Though the difference is more subtle than the 2 aforementioned wiring schemes.

Cool info.

Do you have any wiring diagrams to post?
