I typically run - tuner, wah, compressor, dirt/od, modulation effects (phase, flanger, chorus), delay/reverb in the loop.
I have experimented with phase and flanger into dirt, as well as dirt into wah, and they change it a bit, but I prefered the way I described.

As others have said, I like mine better with my Marshall cab than my Mesa cab. It's a little brighter and open sounding, while still being tight. The Mesa cab is a perhaps a tad bit more tight, but it is also darker and a tad more focused.
