If so How? I mean do you end up running 3 or 4 long cables (I actually use a G5O wireless) >?

I've seen boards set up with a snake going to the amp and then split to front/fx loop for pre and post effects. I think Pete Thorn's new board is set up this way. If long cable runs are a cause for concern, you could use a buffer before hitting your amp, as well one in the loop, if needed.

Flanger, Delay, Tremolo and Reverb all run through my amp's FX loop, but the pedals are located on my pedalboard. I have a 25' snake with midi + three 1/4" runs: one for 'Pedalboard to To Amp Input', one for 'Amp FX Loop Send to Pedalboard' and one for 'Pedalboard to Amp FX Loop Return'. I run a buffer between my guitar and the first pedal on the board, and then also run buffers on the send and return legs of the FX loop. With great results.
