^ You can turn on an amp without a speaker load but you will be in dangerous territory. As long as no signal as sent through the amp it will be fine but generally speaking you want a speaker load (or load dump) on you amp at all times. You can fry an output transformer and power tubes if you don't.
Tap on the preamp tubes with the eraser end of a pencil and listen for the tapping sound coming through the speakers. Like this:
Also, try spraying some contact cleaner on all the tube pins and working the tube in and out of the socket a bit to lubricate and clean.
What mods does the amp have?
Can you try the amp at someone else's house to rule out a dirty power issue?
Also, you can try messing with the hum cancel pot like BrokenFusion recommended but that can be dangerous as you have to take the chassis out. I can help you with that a bit if you are interested.
