The biggest difference I've found is that with the 20th, every control does exactly what it's supposed to and every feature is 100% usable. Whereas the 101b was picky and had tons of "dead spots" in the controls - places that sounded terrible in any application - it truly is difficult to make the 20th sound bad.
That said, they're both fantastic amps. The 20th is without a doubt more versatile, more logical, and easier to use in a live situation, but the 101b is still capable. In terms of sound, the 20th is more thoaty and "on top" of the note. There's less sag than the 101b and when you're really packing on gain with the red channel, it works quite well. The 101b sits a little further "under" the note. It's got quite a bit of sag, but it's also smoother. The red channel is harder to work with because of this, and putting on a ton of gain simply doesn't work as well as with the 20th; it will get muddy.
Red channel - for me - goes to the 20th. The Blue and Green? Green will also go to the 20th. On their own the Greens would be a toss up, but I feel the 20th cleans work better with the other channels; when you play clean for a while and switch to dirt, it sounds fantastic. With the 101b, the Green seems just too different to really fit well. The Blue channel, however, is equally amazing to me on both amps. Again, the difference is between less sag and more upper mids of the 20th to more sag and less upper mids of the 101b.
They do things differently, and the changes are not subtle. If I could only have one (and I only can at this point), it would be the 20th (as it is). Right now, I run everything at noon except volume and it sounds nothing short of staggering. I sold my 101b to get the 20th, and though I would like to own another 101b, I don't regret the decision.
